Uji daya simpan prototype kit vibriosis "RICA"

Abstract
Research on the biomolecular detection of vibriosis have managed to find a specific marker of luminiscent pathogenic Vibrio that the infect specific penaeid shrimp. This specific gene-based marker haemolysin (IAVh). Optimization test indicated that the IAVh primer specific for the detection of vibriosis in shrimp Penaeid with sensitivity is to the density of bacteria 101-102 CFU/mL. The time required for the detection of vibriosis using the AIVh marker is only 1 to 2 days. Furthermore, a prototype detection Kit has been prepared based on PCR method using IAVh primer as its main component and pathogen genomes V. harveyi RICA collection as a positive control. This study aimed to test the storage time of the prototype components Kit vibriosis "RICA". Component Kit consists of two types of mixture. The composition of the first mixture comparises: Taq Buffer dNTP + ddH2O. While the composition of the second mixture comparising a mixture of: Taq DNA Polymerase + buffer + ddH2O. Both kinds of mixed composition are each stored at a temperature of -20oC and -80oC. At the time of PCR process will be implemented, will be added primer and Taq DNA Polymerase for composition 1 and primer and dNTP for composition 2. In the PCR test is used genome of a pure culture of Vibrio pathogens and genomes of diseased organ infection with Vibrio pathogens. The results showed that the composition prototype vibriosis detection kit is the best mixed composition 2 consist of Taq DNA Polymerase + Buffer Taq + ddH2O. The storage temperature is the best: -20oC with optimum storage time for 6 months.
